ABSTRACT

In current study, we aimed to investigate whether the gentiopicroside (GPS) derived from Gentiana manshurica Kitagawa could block the progression of alcoholic hepatic steatosis to fibrosis induced by chronic ethanol intake. C57BL/6 mice were fed an ethanol- containing Lieber-DeCarli diet for 4 weeks. LX-2 human hepatic stellate cells were treated with GPS 1 h prior to transforming growth factor-β (TGF-β) stimulation, and murine hepatocyte AML12 cells were pretreated by GPS 1 h prior to ethanol treatment. GPS inhibited the expression of type I collagen (collagen I), α-smooth muscle actin (α-SMA) and tissue inhibitor of metal protease 1 in ethanol-fed mouse livers with mild fibrosis. In addition, the imbalanced lipid metabolism induced by chronic ethanol-feeding was ameliorated by GPS pretreatment, characterized by the modulation of lipid accumulation. Consistently, GPS inhibited the expression of collagen I and α-SMA in LX-2 cells stimulated by TGF-β. Inhibition of lipid synthesis and promotion of oxidation by GPS were also confirmed in ethanol-treated AML12 cells. GPS could prevent hepatic steatosis advancing to the inception of a mild fibrosis caused by chronic alcohol exposure, suggesting GPS might be a promising therapy for targeting the early stage of alcoholic liver disease.

ABSTRACT

To determine the relationship between the effect of wuzhi capsules on the blood concentration of tacrolimus as compared to diltiazem and with regard to cytochrome P450 (CYP)3A5 gene polymorphisms, 170 patients who underwent renal transplantation from November 2014 to March 2018 and used tacrolimus combined with diltiazem 30 mg bid were selected in this study retrospectively. Patients were divided into an observation group (105 patients) and a control group (65 patients) according to whether they used wuzhi capsules after the operation. The polymorphisms of CYP3A5*3 were determined and the effect of wuzhi capsules on the blood concentration of tacrolimus, as compared with that of diltiazem was determined in patients with different CYP3A5*3 genotypes. This study complies with relevant ethical norms. The results show that compared with diltiazem, an increase of tacrolimus C0/D was significantly correlated with the patient's CYP3A5*3 genotype in both the self-control and the control group. CYP3A5 expressers in the observation group were able to increase the tacrolimus C0/D by about 76.8% by replacing the wuzhi capsules with diltiazem, but this effect was not observed in CYP3A5 non-expressers. In CYP3A5 expressers wuzhi capsules had a greater ability relative to diltiazem to increase the blood concentration of tacrolimus.

ABSTRACT

BACKGROUND@#Lung cancer is the cancer with the highest morbidity and mortality at home and abroad at present. Using computed tomography (CT) to screen lung cancer nodules is a huge workload. To test the effect of artificial intelligence in automatic identification of lung cancer by using artificial intelligence to find the lung cancer nodules automatically in the chest CT of 1 mm and 5 mm thick.@*METHODS@#5,000 cases of T1 stage lung cancer patients with 1 mm and 5 mm layer thickness were respectively labeled and learned by computer neural network, the algorithm of forming pulmonary nodules was carried out. 500 cases of chest CT in T1 stage lung cancer patients with 1 mm and 5 mm thickness were tested by artificial intelligence formation, and the sensitivity and specificity were compared with artificial reading.@*RESULTS@#Using artificial intelligence to read chest CT 500 in 5 mm, the sensitivity was 95.20%, the specificity was 93.20%, and the Kappa value of two times repeated read was 0.926,1. For 1 mm chest CT 500 cases, the sensitivity is 96.40%, the specificity is 95.60%, and the Kappa reads two times is 0.938,6. Compared with 5 doctors, the same CT sets with 1 mm thickness were read. The detection rates of artificial intelligence and artificial reading were similar to those of lung cancer nodules and negative control read films, and there was no significant difference between them. In the comparison of the same CT slices with 5 mm thickness, the number of detection of lung cancer nodules by artificial intelligence is better than that of artificial reading, and the sensitivity is higher, but the number of false messages is increased and the specificity is slightly worse.@*CONCLUSIONS@#The automatic learning of early lung cancer chest CT images by artificial intelligence can achieve high sensitivity and specificity of early lung cancer identification, and assist doctors in the diagnosis of lung cancer.

ABSTRACT

Objective: To detect the expressions of periostin, matrix metalloproteinase-8 (MMP-8) and tissue inhibitor of matrix metalloproteinase-2 (TIMP-2) in gingival tissue of the patients with chronic periodontitis, and to explore the possible role of periostin in the occurrence and development of chronic periodontitis and its related biological mechanism. Methods: A total of 23 normal gingival tissues (control group) and 19 inflammatory gingival tissues (case group) were selected. The expressions of periostin, MMP-8 and TIMP-2 in gingival tissue of the patients in two groups were detected by immunohistochemical SP method. The correlation between the expressions of periositn and MMP-8 and TIMP-2 in gingival tissue of the patients in two groups were detected by Spearman analysis. Results: The positive expression rate of periostin in gingival tissue of the patients in case group was lower than that in control group (P<0. 05). The correlative analysis results demonstrated that the expression of periostin had negative relationship with the expression of MMP-8 (r=-0.51, P<0. 05) in control group, and positive relationship with the expression of TIMP-2 (r=0. 50, P<0. 05); in case group, the expression of periostin had negative relationship with the expression of MMP-8 (r=-0.482, P<0. 05), and positive relationship with the expression of TIMP-2 (r= 0.655, P<0. 05). Conclusion: Periostin can be used as a preliminary indicator for maintaining the structural stability of gingival tissue, and become a research target for periodontal tissue regeneration.

ABSTRACT

·AlM:To study the MRG15 (death factor related gene) in age-related cataract ( ARC ) , differential expression of normal lens epithelial cells. · METHODS: Forty mature healthy female SD rats were randomly divided into study group and control group, 20 rats in each group, the study group were ovariectomized, low concentration of naphthalene long interval of administration, the establishment of perimenopausal ARC model, the control group of conventional farming.ln the subtractive hybridization cloning by MRG15, and make the probe of the cDNA fragment using digoxigenin labeled. Access to the two groups of rats anterior lens capsule after slicing, and then through the differential expression in situ hybridization clear lens epithelial cells. · RESULTS: The cloned MRG15 through BamH1, EcoR1 enzyme digestion and agarose gel electrophoresis, available for 639bp long cDNA fragment. GeneBank display contrast, their homology was 99.0%. ln situ hybridization, ARC patients and normal lens epithelial cells were observed in the expression of MRG15.Study group the percentage of positive cells compared with control group, showed a significant difference (P<0.05). lntegral index study group and the control group compared with, was significantly difference (P<0.05). · CONCLUSlON: ARC, MRG15 in normal lens epithelial cells expressed ARC, and compared with the normal expression of lens epithelial cells, which may produce inhibitory effects are associated with MRG15 transcription in human lens epithelial cells in the part of key genes, by reducing the lens epithelial cell fu nction, make its appear aging, and the formation of cataract, clinical response to this should further study.
